Polarized cells display asymmetric release of extracellular vesicles

چکیده

Extracellular vesicles (EVs), a broad term for the lipid microparticles known as microvesicles and exosomes, are discharged by cells into their surrounding space. Microvesicles upon outward plasma membrane budding, while exosomes secreted after multivesicular body (MVB) fusion with membrane. The majority of information regarding EV biology comes from studies performed in non-polarized cells. Here we characterize release polarized We found substantial asymmetry number composition EVs produced released apical epithelial compared to basolateral showed that quantitative difference is related distribution two phosphoinositide species between cell surfaces peculiar biochemical resultant reflects site origin. In particular, may derive distinct classes MVBs originating fusing same identify VAMP8/Endobrevin regulator whereas mechanism responsible requires further study.

Extracellular Vesicles from Ovarian Carcinoma Cells Display Specific Glycosignatures

Cells release vesicles to the extracellular environment with characteristic nucleic acid, protein, lipid, and glycan composition. Here we have isolated and characterized extracellular vesicles (EVs) and total cell membranes (MBs) from ovarian carcinoma OVMz cells. EVs were enriched in specific markers, including Tsg101, CD63, CD9, annexin-I, and MBs contained markers of cellular membrane compar...

Polarized Cells, Polarized Views: Asymmetric Cell Division in Hematopoietic Cells

It has long been recognized that alterations in cell shape and polarity play important roles in coordinating lymphocyte functions. In the last decade, a new aspect of lymphocyte polarity has attracted much attention, termed asymmetric cell division (ACD). ACD has previously been shown to dictate or influence many aspects of development in model organisms such as the worm and the fly, and to be ...

Selective release of circRNAs in platelet-derived extracellular vesicles

Circular RNAs (circRNAs) are a novel class of noncoding RNAs present in all eukaryotic cells investigated so far and generated by a special mode of alternative splicing of pre-mRNAs. Thereby, single exons, or multiple adjacent and spliced exons, are released in a circular form. CircRNAs are cell-type specifically expressed, are unusually stable, and can be found in various body fluids such as b...

Microbubbles-Assisted Ultrasound Triggers the Release of Extracellular Vesicles

Microbubbles-assisted ultrasound (USMB) has shown promise in improving local drug delivery. The formation of transient membrane pores and endocytosis are reported to be enhanced by USMB, and they contribute to cellular drug uptake. Exocytosis also seems to be linked to endocytosis upon USMB treatment. Based on this rationale, we investigated whether USMB triggers exocytosis resulting in the rel...

Isolation and characterization of Faecalibacterium prausnitzii extracellular vesicles

Introduction: Extracellular vesicles (EVs) contain active biological compounds which play important roles in biological processes. The secretion of EVs is a common phenomenon which occurs in archaea, bacteria and mammalian cells. The secretion of bacterial EVs has been discovered in various species of both Gram-negative and Gram-positive bacteria. Faecalibacterium prausnitzii is one of the comm...
